Flow Cytometric Analysis There are at least two gating strategies that may be applied for flow cytometric analysis of mtracellular cytokines. They are conventional FWD vs SSC (light scatter) gating and immunoscatter gating, in which one of the fluorescence parameters is plotted against the SSC signal. Examples of both types of analysis strategy are demonstrated in Fig. 1. Light-scatter gating works well with short-term activated cells m which the light-scatter properties have not been altered significantly (for example, PBMCs cultured for 4 h with PMA + ionomycin).
