The process of opening ampoules and recovering freeze dried cultures can be difficult.
The sterile loop is dipped into the sample from which the organism is to be isolated, and is then drawn gently across the surface of the agar several times to cover an arc of about sixty degrees. The loop is resterilised in the bunsen flame or in the case of plastic loops, a fresh one is used. The loop is then drawn through the tail end of the previous streaks across a second arc of sixty degrees. This is repeated a third and fourth time. 1).
